Measure EV Purity Results by Calculating the Membrane Ratio with the Exoplorer

Extracellular Vesicles (EVs) quality plays a crucial role in ensuring the reliability of research and the effectiveness of therapeutic applications. Low-purity samples can lead to misleading results and wasted resources — that’s why having a fast, accurate way to measure EV quality is essential. To prove its performance, comparative tests were conducted between Exoplorer™ and the commonly used Nanoparticle Tracking Analysis (NTA). Two recognized approaches were used to assess purity: the particle/protein ratio and the membrane ratio. Both methods confirmed that Exoplorer™ provides accurate, reproducible results, clearly distinguishing between high-purity and lower-purity EV samples.
Using the particle/protein ratio — a method recommended by the International Society for Extracellular Vesicles — Exoplorer™ matched or exceeded NTA in detecting EV concentration. Protein content was determined via Bicinchoninic Acid Assay, and the calculated ratios confirmed the high sensitivity of Exoplorer™ in identifying sample purity. Similarly, the membrane ratio method, based on measuring intact vesicles before and after lysis, showed that Exoplorer™ is highly capable of quantifying EVs with precision.

In a comparison of two commercial EV samples — 293F-exo (high purity) and MSC-exo (lower purity) — Exoplorer™ delivered clear and reliable results.
