Spatially Resolved Proteomic Profiling of Nuclear and Cytoplasmic Compartments using Microscoop®

The functional organization of the eukaryotic cell is defined by its precise compartmentalization, in which distinct biological processes are spatially segregated across organelles to maintain metabolic efficiency and regulatory control. Accurate characterization of compartment-specific proteomes is therefore critical for understanding cellular homeostasis, signaling dynamics, and disease mechanisms. However, conventional approaches for subcellular proteome analysis, such as biochemical fractionation, often suffer from limited spatial resolution and cross-contamination between compartments. Proximity labeling (PL) techniques, while powerful, can be constrained by the requirement for specific bait proteins, the need for fresh or living samples to enable enzymatic labeling and the potential diffusion of reactive labeling species, complicating the assignment of proteins to their true subcellular origins. Together, these limitations hinder confident discrimination between true compartment of localized proteins and experimental artifacts.

To address the need for high-resolution proteomic characterization from preserved native cellular architecture and protein localization, Syncell introduced the Microscoop platform, a spatial proteomics technology that integrates microscopy with targeted photolabeling. Through microscopy-guided region-of-interest (ROI) selection, Microscoop enables sub-micron targeting of selected regions and localized biotinylation of proteins, facilitating accurate proteome characterization directly within intact biological specimens.
