Key Methodology: Organ In Vivo Imaging by Lago
The Lago X (Spectral Instruments Imaging) system was utilized for high-sensitivity optical in vivo imaging to track the systemic distribution and efficacy of the nanoparticles.
Method: Mice were injected with nanoparticles (M30 D90) carrying a luciferase reporter plasmid (pLuc). After 24–48 hours, the substrate D-luciferin was administered, and the Lago system captured bioluminescence signals from the whole body and specific isolated organs.
Detection: The Lago system’s high-throughput capability allowed researchers to quantify exactly where the gene expression (transfection) occurred. It provided real-time, non-invasive visualization of luciferase activity, which was then validated by ex vivo imaging of harvested organs (brain, liver, lungs, spleen, and kidneys).
Key Findings
Caveolae Induction: Systemic mannitol administration at a specific dose (3 g/kg) does not just “shrink” cells to open tight junctions; it actively increases the abundance of caveolae (small pits/vesicles) in the brain’s endothelial cells.
Targeted Delivery: The mannitol-modified nanoparticles (M30 D90) specifically exploit these induced caveolae. This resulted in a significant increase in gene delivery to the brain compared to standard nanoparticles.
Liver Evasion: A major breakthrough was the reduction of “off-target” effects. While many nanoparticles get trapped in the liver, the M30 D90 particles showed minimal liver accumulation, prioritizing brain uptake.
Neuronal Transfection: Once across the BBB, the nanoparticles successfully transfected neuronal cells, demonstrating the platform’s potential for treating neurological disorders.
How Organ Imaging Helped the Study
Verification of the BBB Crossing: Without organ imaging, it would be impossible to prove that the bioluminescence was coming from inside the brain tissue rather than the surrounding vasculature.
Biodistribution Mapping: The Lago imaging provided a comparative “heat map” showing that the M30 D90 nanoparticles were superior to the control (D90) in avoiding the liver and lungs while maximizing brain signal.
Dose Optimization: It allowed researchers to determine the optimal timing and concentration of mannitol needed to trigger the caveolae-mediated pathway without causing systemic inflammation or permanent BBB damage.
